I am interested in the  regions of the brain that correlate with hippocampal activation when using a spatial strategy during navigation. During a block I have multiple conditions, but I am really only interested in my test condition. Can I run a PLS analysis if I am only intersted in correlations for a single condition? If I want to know what regions correlate with my hippocampal seed during a single condition, what might the best approach be for this analysis?

You can indeed run the analysis with one conditon, but you should mindful of what conclusions you will be able to draw.  By analogy, consider activation analysis where you have only one condition.  You can make some statement about the pattern of activity but not whether it differs between tasks.  For correlation, you could say that hippocampus is significantly correlated with a specific pattern in the navigation task, but can't infer whether that is unique to the task or sort of "obligatory" connectivity that is always there.
